The Research Assistant will take part in ongoing work funded by the . This project seeks to address the important and challenging task of developing new microbial hosts for biofuel production. The Research Assistant will directly contribute to bench-level experimental research involved in microbial strain engineering and optimization to unlock the potential of renewable energy and other biologically derived materials from single cell organisms, such as Pseudomonas putida. What You Will Do: DNA design, manipulation, and cloning. Engineering of bacterial strains such as Pseudomonas putida, Corynebacterium glutamicum, and Escherichia coli with classical and next generation editing methods, such as homologous recombination and CRISPR. Functional expression and evaluation of candidate pathway genes in microbial hosts, especially in the context of Sartorius 2L bioreactor scale-up systems (and similar). Growing, manipulating and archiving bacterial cultures. Metabolite and proteomic sample prep for LC- and GC-MS. Additional Responsibilities as needed: Data recording and report preparation for supervising scientists. Participating in community laboratory jobs at the RA level. (preparation of antibiotics, buffer/media preparation) Miscellaneous tasks required to ensure a functional research team. (preparing competent cells, ordering supplies, etc.) What is Required: B.S. degree in microbial or molecular biology preferred with demonstrated success in applying molecular biology research techniques. Working experience in a fast-paced biotechnology startup environment. (preferably at least six months) Experience in the following techniques: PCR; DNA sequence analysis; DNA isolation, gene cloning and expression; genetic manipulation and cultivation of E. coli or Pseudomonas putida. Familiarity with recent cloning methods. (Gibson, HiFi assembly, Cas9/CRISPR etc.) Strong analytical and communication skills. Ability to be cooperative, team-oriented, mission-oriented, and maintain an up-to-date lab notebook. Must adhere to all campus and EH&S safety policies and model safe lab best practices for others. Additional Desired Qualifications: Experience with bacterial strain growth and genetics in Pseudomonas putida Familiarity with Sartorius Biostat B bioreactors or Sartorius Ambr250 automated bioreactors Familiarity with ssDNA oligo recombineering a plus. The posting shall remain open until the position is filled. Notes: This is a full time, 1-year term appointment with the possibility of extension or conversion to Career appointment based upon satisfactory job performance, continuing availability of funds and ongoing operational needs. M-F, Non-exempt (hourly paid) from overtime pay. This position is represented by a union for collective bargaining purposes.
